Quick Transformation:

 

Thaw competent cells on ice.

Chill eppendorff tubes and electroporation cuvettes.

Warm SOC media to RT.

Set volts on electroporator to 2000V.

 

  1. In an eppendorff tube combine 20ul competent cells and 0.2-2ul DNA. Be gentle and do not pipette up and down.
  2. Transfer into electroporation cuvette.
  3. Place cuvette in electroporator and zap (press button twice).
  4. If you here a POP there is too much DNA, repeat with less DNA.
  5. Add 200ul SOC media and place in eppendorff
  6. incubate at 37C with shaking 250rpm for 30-60min
  7. Plate out on LB plates containing the suitable antibiotic, 37C O/N

 

Wrap plates in parafilm and store at 4C

 

Minipreps:

 

In the afternoon pick individual colonies into 5ml LB with antibiotic and grow up O/N @37C, 250rpm.

 

Midipreps:

 

In the morning pick individual colonies in to 5ml LB with antibiotic and grow up during the day @37C, 250rpm. Transfer into 50-100ml LB with antibiotic and grow up O/N @ 37C, 250rpm.
